This Homogeneous TRF-PINCER® Assay is intended for quick screening and quantification of Biotin in biological samples.
Biotin, also known as vitamin H or B7, is a water-soluble B-complex vitamin which plays an indispensable role in numerous naturally occurring carboxylation reactions in living cells. Biotin is also very commonly used to label peptides, DNA or proteins. The strong binding between biotin and the tetrameric protein avidin, streptavidin, or neutravidin (with a dissociation constant (Kd) in the order of 10-14 M) allow biotin to be used in many different biotechnological applications. The Biotin PINCER® Assay Kit provides a convenient method to quantify protein concentration in unknown solution as well as estimate the amount of biotin in biotinylated samples.

Principle of the Assay
Two biotinylated PINCERS have oligonucleotides (with short complementary sequences) labeled with fluorescent probes. The PINCERS will form a complex upon binding to streptavidin and activate Time-Resolved Fluorescence FRET (fluorescence resonance energy transfer).
The fluorescence signals can be customized to fit within an appropriate measurable range by adding non-fluorescently labeled PINCERS. When non-fluorescently labeled biotinylated PINCERS are present, they will compete with the fluorescence-labeled biotinylated PINCER and result in a decreased TRF-FRET signal (see figure below).
